Processing Instructions

CSF should not be haemolysed or xanthochromic and must contain less than 500 RBC and <= 10 WBC per uL. Cell count must be performed on the tube to be sent (or a lower numbered tube). Send to Lab before 4pm.

CSF should be transferred (unspun) into a labelled screw top container using a sterile pipette as soon as possible after collection. Parafilm the lid. Initial label to indicate patient details have been checked.

Freeze specimen at -20'C and store frozen. Record tube used and the date and time of freezing on the request form.

FNA Specimen Collection Using Cytolyt (LBC) - For Cytology Tests

Please note: CytoLyt solution is not a suitable medium for the collection of specimens for Flow cytometry, Microbiology, and Biochemistry testing. Please check other laboratory collection requirements.

Collection Containers -  FNA Kit (containing slides, slide mailers and Cytolyt Pots) are available from NPV Anatomical Pathology.
• Specimen Pot (CytoLyt) -  label the pot with the Patients Name, DOB and Specimen type/site.
• Slides - Label the frosted end of 2 slides with Patients Name, DOB and Specimen type/site.

1. After the first pass of the specimen has been aspirated, immediately express a small amount of FNA specimen on to one of the microscope slides and smear gently with the other labelled slide. Allow these to air dry.
2. Express the remaining FNA material into the labelled pot containing Cytolyt.
3. Rinse the needle by taking up the CytoLyt solution into the needle and expressing it .This can be performed a number of times to ensure all material from the needle and syringe has been retrieved.
4. Further passes from the same site can be collected with a new needle and aspirated into the original pot, no further slides are required to be prepared.
5. If several different sites are to be sampled, please use a separate FNA kit, containing a pot of CytoLyt for each site/lesion sampled. Making 2 smears for only for the first pass.

6. Place the specimen pot(s) and slide mailers (containing the slides), in abiohazard bag for transport to Anatomical Pathology.

Frozen Sections

Frozen sections can only be performed on fresh unfixed tissue

The Anatomical Pathology Unit should be given 24 hours notice of a frozen section to ensure a Consultant Pathologist will be available when the frozen section arrives (telephone 9485 9333).

A frozen section must not be sent to the laboratory unannounced

Infectious Specimens: If there is a danger of infection (eg tuberculosis, hepatitis, CJD etc) please contact a pathologist before the procedure to discuss alternate methods for  rapid diagnosis.

For a frozen section the fresh tissue is sent to the Anatomical Pathology Unit in a labelled container with an accompanying request form https://northernpath.org.au/wp-content/uploads/2024/02/Frozen-Section-Booking-Form-blank-p6-003.pdf . The request form should note the clinical and operative findings.

Samples suitable for auto-antibody screening:

Blood Serum: request for 5.0 mls of blood collected in serum gel tube (SST).

CSF: request for 1.0ml

Samples not available for delivery following collection are to be stored at 4o C. Storage at 4oC is suitable for up to 2 weeks thereafter samples need to be frozen and sent on dry ice.

Muscle Biopsy

Muscle biopsy for neurodegenerative and metabolic studies are processed by the Victorian Neuromuscular Laboratory Service (VNLS) which is located within the Anatomical Pathology Department of AlfredHealth.

Important Note To Clinicians: The Neuromuscular laboratory must receive samples BEFORE 4pm Monday to Friday to enable optimal handling and processing to occur.  Clinicians must allow sufficient transport time when scheduling procedures.

Muscle biopsy Kits are unavailable from VNLS.

Muscle Biopsie Procedure.

Correct handling of a muscle biopsy is important for interpretation of the specimen.

  • Please deliver speciment to NPV lab by 11:00am.
  • Please do not freeze the specimen.
  • Do not immerse the specimen in any fluid.
  • Do not use any diathermy instrument on the muscle specimen.
  • do not infuse the muscle with local anesthetic.

One (1) specimen is required.

A 20 x 10 x 10mm rectangular piece of muscle is collected from the patient and placed in a dry sterile container (70ml Yellow top container).

Place this specimen in an esky with ice cubes and send immediately to NPV lab.

Specimen Dispatch

Muscle biopsies and completed paperwork should be directed to Anatomical Pathology via Central Specimen Processing.
