FNA Specimen Collection Using Cytolyt (LBC) - For Cytology Tests

Please note: CytoLyt solution is not a suitable medium for the collection of specimens for Flow cytometry, Microbiology, and Biochemistry testing. Please check other laboratory collection requirements.

Collection Containers -  FNA Kit (containing slides, slide mailers and Cytolyt Pots) are available from NPV Anatomical Pathology.
• Specimen Pot (CytoLyt) -  label the pot with the Patients Name, DOB and Specimen type/site.
• Slides - Label the frosted end of 2 slides with Patients Name, DOB and Specimen type/site.

1. After the first pass of the specimen has been aspirated, immediately express a small amount of FNA specimen on to one of the microscope slides and smear gently with the other labelled slide. Allow these to air dry.
2. Express the remaining FNA material into the labelled pot containing Cytolyt.
3. Rinse the needle by taking up the CytoLyt solution into the needle and expressing it .This can be performed a number of times to ensure all material from the needle and syringe has been retrieved.
4. Further passes from the same site can be collected with a new needle and aspirated into the original pot, no further slides are required to be prepared.
5. If several different sites are to be sampled, please use a separate FNA kit, containing a pot of CytoLyt for each site/lesion sampled. Making 2 smears for only for the first pass.

6. Place the specimen pot(s) and slide mailers (containing the slides), in abiohazard bag for transport to Anatomical Pathology.

If the sample is not collected at Northern Hospital Epping i.e. collected at other collection sites (Broadmeadows Hospital, Bundoora Centre and Craigieburn Centre), the primary SST tube shall be centrifuged and the sample kept cool with ice blocks at all times. When the SST sample arrives at the laboratory, aliquot and freeze serum at -20 degrees Celsius.

Guidelines for swab collection.

A sterile flocked swab is used to vigorously swab the base of the lesion— applying enough pressure to collect epithelial cells without causing bleeding—and collect vesicular fluid. It is important to collect infected epithelial cells from the base of the lesion because they usually contain a significant amount of virus. 
Break off swab into red top swab container.

For investigation of CMV colitis, a colonic biopsy is recommended. For all other enquires, please contact the Microbiology Registrar.

Please provide relevant clinical history and specify exact type of flow cytometric analysis required.

For peripheral blood samplean FBE must also be requested. Collect 2xEDTA. 

For CSF sample, please collect a tube with 2-3mL RPMI from Central Specimen Reception BEFORE the CSF sample is collected. CSF should be collected directly into this tube (1 part RPMI; 1 part CSF) to improve sample yield. All CSF flow requests must also have CYTOLOGY requested.

Samples must be marked “Urgent”. Flow cytometry is NOT available after 1pm on Fridays or during weekends/afterhours. Testing during these times require approval from a laboratory haematologist PRIOR to sample collection. Otherwise, the sample may not be tested in time (within 2-4 hours for CSF, up to 24 hours for blood sample) or tested at all. Samples not processed in time is non-viable and non-diagnostic. 

Laboratory haematologist can be contacted on 8405 2596 during normal working hours or via switchboard after hours.

For CSF sample, please collect a tube with 2-3mL RPMI from Central Specimen Reception BEFORE the CSF sample is collected. CSF should be collected directly into this tube (1 part RPMI; 1 part CSF) to improve sample yield. All CSF flow requests must also have CYTOLOGY requested.

Samples must be marked “Urgent”. Flow cytometry is NOT available after 1pm on Fridays or during weekends/afterhours. Testing during these times require approval from a laboratory haematologist PRIOR to sample collection. Otherwise, the sample may not be tested in time (within 2-4 hours for CSF, up to 24 hours for blood sample) or tested at all. Samples not processed in time is non-viable and non-diagnostic.

Microarray provides a superior investigation than FISH testing as it detect a wider range of genetic changes that may cause each syndrome. Registration Instructions

2x EDTA lavender top tube required for thalassaemia testing (1x Hb Electophoresis, 1x DNA testing if required)

1x Serum gold top tube

Please register THL, FBE and FER panel when for all thalassaemia screens.

DO NOT SEND TO MONASH DIRECTLY FROM CSR. If the word gene or DNA is used in the request, this is for genetic test. Thalassaemia genetic test is performed at Monash, register as MON. ALL Thalassaemia DNA analysis must be approved by the Haematology Scientist, Registrar or Consultant first. DO NOT SEND TO MONASH DIRECTLY FROM CSR. All samples requiring DNA analysis will be packaged up by the scientist prior to handing over to the sendaway department for sendout. 

FBE, ferritin and Hb electrophoresis results must be sent together with second EDTA tube.

Patient needs to provided Australian Medicare number and Victoria address for state goverment grant (No charge). For Overseas residents with Victorian address a bill will be issued.
